個人簡介

方沈應,南方科技大學公共衛(wèi)生及應急管理學院教授、研究員、博士生導師。獲美國德克薩斯大學生物統(tǒng)計學博士,曾任德克薩斯大學MD安德森癌癥研究中心腫瘤外科系助理教授、副教授,武漢科技大學特聘教授,廣州醫(yī)科大學“南山學者”特聘教授。任PLOS ONE、Advances in Public Health 及 Frontiers in Oncology雜志編輯,擔任American Journal of Human Genetics、Journal of National Cancer Institute、PLOS Genetics、International Journal of Cancer、Carcinogenesis以及Scientific Reports等13本英文雜志評委,并參與National Science Center (NCN panel NZ7, Poland)和中國國家自然基金委基金評審;為美國癌癥協(xié)會、遺傳學會和統(tǒng)計學會會員。主要運用流行病學與生物統(tǒng)計學方法研究環(huán)境因素、遺傳因素與惡性腫瘤或慢性病的發(fā)生、發(fā)展及預后的關系。參與收集了北美最大的黑色素瘤病例對照研究人群并創(chuàng)建了北美黑色素瘤風險預測模型。目前主要運用遺傳學、生物信息學及機器學習法,深度挖掘研究病人個體基因組學特征、疾病臨床表現(xiàn)信息,包括影像學資料和病理組織學染色圖片等醫(yī)療大數據,為腫瘤的早期診斷、個體化治療、疾病復發(fā)及生存預后提供相應依據。研究成果發(fā)表于J Clin Oncol.(第一作者3篇, 共同作者5篇), J Invest Dermatol.(通訊作者4篇), Nature Genetics(4篇),Int J Cancer(第一作者3篇, 共同作者6篇),The Lancet Oncology(1篇),Cancer(6篇)等國際著名刊物。近期的研究成果被Cancer network、NEJM等專業(yè)媒體報道, 于2011美國遺傳學年會及2020年全球華人統(tǒng)計學年會獲大會報告, 在SCI等刊物發(fā)表英文論文70余篇。


研究方向

1. 人群流行病學研究:依據大灣區(qū)人群樣本,開展人群流行病學觀察研究,調查研究環(huán)境因素與遺傳因素在慢性病、腫瘤或新發(fā)傳染病發(fā)生、發(fā)展中的作用,為采取干預措施以降低疾病的發(fā)病風險提供依據。

2. 健康醫(yī)療大數據應用:(1)依托附屬醫(yī)院及合作醫(yī)療單位,收集腫瘤病人的血液及組織樣本,建立患者醫(yī)療數據庫;(2)結合TCGA腫瘤數據庫及本地醫(yī)院人群標本庫,同MD安德森研究中心、Baylor醫(yī)學院以及St Jude兒童醫(yī)院等開展多中心合作,整合分析多組學—基因組、轉錄組、表觀基因組、蛋白組及代謝組學等多層組學數據,同時納入人口統(tǒng)計學或臨床表型變量,應用統(tǒng)計學與人工智能新理論—神經網絡模型(CNN)、變分自編碼器模型(VAE)以及條件生成對抗網絡模型(CGAN),對高維數據降維并壓縮提取特征變量,以預測疾病的發(fā)生、發(fā)展及預后,并為精準治療提供依據;(3)機器學習及數據科學在醫(yī)療影像資料—數字化組織病理學圖像、CT、MRI等中的應用,包括質量控制、聚類分割、標識、特征提取及結果預測,為提高疾病的診斷、危險度分層及指導精準治療和預后評估提供依據,并且可藉此發(fā)展出新的治療手段與科技轉化平臺;發(fā)展機器學習新模型-Transformer,并應用于圖像處理分析以及識別腫瘤驅動基因;(4)協(xié)同附屬醫(yī)院,開展臨床流行病學研究,科學實施臨床實驗設計與診斷性試驗評價,以及針對治療方案開展衛(wèi)生經濟學評估與決策分析,為政策制訂者提供依據。

3. 生物信息學研究:評估腫瘤微環(huán)境中的免疫細胞、腫瘤細胞及間質細胞的構成演變在腫瘤轉移、預后與治療中的作用。

4. 疾病病因學推論:孟德爾隨機化研究設計及病因推論;運用加性噪聲模型進行因果關聯(lián)分析。

5. 統(tǒng)計學模型在遺傳流行病學中的應用:多因子風險評估、基因-基因,基因-環(huán)境交互作用。
